    1979 7 1 斯坦福大学 George Stark 实验室发表于《美国国家科学院院刊》(PNAS)的论文:Transfer of proteins from gels to diazobenzyloxymethyl-paper and detection with antisera: a method for studying antibody specificity and antigen structure. (1979) PNAS 76:3116-3120. 这篇文章17974月份提交,19797月份发表。他们但是用的是I125标记的链球菌protein A作为二抗探针, Western Blot结果是长这样:

    ·      同样是在1979年,也就是晚了那么一两个月,瑞士巴塞尔的Towbin研究团队,首先用电转移的方法将SDS-PAGE上的蛋白转移到膜上,替代了Stark的被动转印,与我们现在的Western blot模式基本相同;


    同样是1979,工作于西雅图弗雷德·哈钦森癌症研究中心Burnette也发明这一技术,但是他的文章直到1981年才发表,这个家伙把这个技术定义为“Western Blot”以区别于Southern blot,因为他们实验室正好也处于美国西海岸。Burnette独立开发了他的这个技术,尤其是电泳欢饮步骤,但是在1979年他提交他的论文之前,他已经看过了Stark Towbin技术文献,但是他坚信自己的技术更简单方便,应该北发表;但是他的论文被拒,虽然他的技术不是在Towbin的论文基础上改造的,但是显然两个技术明显相同,不但论文没有北发表,很多人还嘲笑蹭Edwin Mellor Southern Southern blot的热度,你以为你的发明在西海岸,你就命名为“western Blot”。既然这个技术不能北公开发表,Burnette把这个技术方案复印了很多份传给朋友们,很快就开了一个关于blotting技术的电话研讨会,他需要想每一个人解释这个技术,因为很多人拿到复印件之后可能根本读不懂; 那段时间Burnette 几乎苏哦有的时间都在打电话解释他的[Western blotting]技术。通过民间传播这个技术声名远扬,他的文章终于也在1981年北公开发表。

    这个篇文章题目是: "Western blotting": electrophoretic transfer of proteins from sodium dodecyl sulfate-polyacrylamide gels to unmodified nitrocellulose and radiographic detection with antibody and radioiodinated protein A. (1981) Analytical Biochemistry 112:195-203. 


    那么究竟这个western Blot技术的功劳归谁呢?我们觉得他们都是功不可没。

     

    1979年注定是不平凡的一年,所谓英雄所见略同,文中提到的大牛George Stark还在1975年发明了Northern blot技术,直到现在他的身影还活跃在科研一线,广东海大和青岛海洋研究所都跟他有密切合作关系;除此之外,还是在1975年,文中提到的Edwin Mellor Southern,这个英国人发明了Southern blot技术。科学的故事总是充满趣味的,科学研究过程往往是枯燥无味。
